Xenon Pharmaceuticals paused enrollment in its major depressive disorder and bipolar depression trials after unexpected neuropsychiatric adverse events emerged. The halt underscores the safety challenges inherent in psychiatric drug development, where unexpected side effects can derail promising candidates.

In contrast, other treatments advanced. Johnson & Johnson's lumateperone met its phase III primary endpoint, rapidly reducing acute manic symptoms in bipolar I disorder as early as day 3. Alkermes reported positive phase Ib proof-of-concept data for its investigational orexin 2 receptor agonist, showing clinically meaningful ADHD symptom reduction in adults. Otsuka's centanafadine also demonstrated consistent improvements across ADHD and anxiety symptoms, emotional dysregulation, and executive function in a phase IIIb trial.

Meanwhile, CDC data from 2023 to 2025 showed improvements in many mental health and suicide risk indicators among youth, and a cross-sectional study found Gen Z and Millennials drove recent drops in alcohol use. However, a U.K. analysis identified children as young as 13 presenting with ketamine-induced uropathy, highlighting ongoing substance-related risks.

Experts also advised psychiatrists to explicitly define near-term treatment goals when prescribing benzodiazepines and to gain competency in deaf and hard-of-hearing care to avoid misinterpreting patient expression and thought processes.
